Gene silencing is a natural cellular phenomenon, implements blocking by causing degradation of the intermediate RNA form before the RNA code can be translated into a protein. The most commonly used methods of gene silencing are RNA interference (RNAi) and antisense oligonucleotide (ASO) technologies. These two methods have proven their potentials for gene therapy applications, offering great promise for curing diseases and drug research.

Genetic therapies can be separated into three types, namely short synthetic nucleotides, virus-mediated gene therapy and genomic DNA editing/engineering. RNAi and ASOs are the examples of short synthetic nucleotides. RNAi-based and ASOs-based therapies are synthetic strands of nucleotides bind individual RNAs through canonical Watson-Crick base pairing. However, these two methods have different mechanisms. The mechanisms of action for ASOs are through degrading RNA, blocking protein translation and modifying RNA splicing. As to the RNAi-based therapy, it works through the RNA-induced silencing complex (RISC)-mediated cleavage of messenger RNA to prevent the formation of the proteins that they code for.
