RNA interference (RNAi) is a biological procedure that gene expression or translation is restrained by targeted mRNA molecules. The mechanism of RNAi includes initiation and effector steps. Firstly, the dsRNA is recognized and cleaved into siRNAs with a length of 21-23 nucleotides by Dicer. Then these siRNAs are integrated into a RNA-induced silencing complex (RISC), which acts on the cognate mRNA and degrades it. The RNAi mechanism for sequence-specific gene silencing has essential applications in therapeutic intervention and target identification of drug discovery. RNAi has been regarded as a therapeutic method for disease treatment, such as the treatment of age-related macular degeneration.
